I tried searching but couldn't find a clear answer. I'm planning on ordering wheels for my 2017 TLX in 19x8.5 with a +40 offset. Tire size will be 245/40/19.

Any issues with fitment here on stock suspension. Will I need to roll the fenders or will it fit completely stock?

You'll be fine. I have 245/35/20 on my 2017 TLX and they are virtually the same size as you are considering. I'll be putting them on my new 20 A-spec I leased today. Also, just check tire size.com for reference.

I am running the Acura OEM 19" rims on my car with the 245/40/19 tires.

Thanks! What the size and offset on your wheels? Whats the make and model of the wheels?

Thanks! These are +50 offset, just the factory wheels.
